Meloxicam tablets should not be used in children who weigh less than 60 kg (132 lbs). Given the disparity in sex distribution across study groups, an analysis by sex was conducted. No significant differences in any efficacy parameter were found between male and female subjects.
Side effects of NSAIDs
In published animal studies, prostaglandin synthesis inhibitors have been reported to impair kidney development when administered at clinically relevant doses. In animal reproduction studies, embryofetal death was observed in rats and rabbits treated during the period of organogenesis with meloxicam at oral doses equivalent to 0.65- and 6.5-times the maximum recommended human dose (MRHD) of meloxicam. Increased incidence of septal heart defects were observed in rabbits treated throughout embryogenesis with meloxicam at an oral dose equivalent to 78-times the MRHD. In pre- and post-natal reproduction studies, there was an increased incidence of dystocia, delayed parturition, and decreased offspring survival at 0.08-times MRHD of meloxicam.

Infusion‐site extravasation was reported for 1 patient in the ibuprofen group, and injection‐site pain was reported for 1 patient in the placebo group. Combining alcohol with either meloxicam or ibuprofen can be dangerous. However, some authors hypothesized that kidney outcomes of NSAIDs were not dependent on COX-2 selectivity (43). Instead, physiochemical properties that determine drug distribution in the kidney relative to plasma concentration play a more important role, rendering celecoxib more nephrotoxic than meloxicam, despite both of them being COX-2 selective (43,44). Nonetheless, this study was unable to validate this hypothesis because meloxicam was not included. Instead of being two distinct drug moieties, COX-2 selective inhibitors and other NSAIDs have different positions along the continuous scale of COX selectivity, in addition to their different potencies and pharmacokinetics properties (42). Some investigators also suspected that differences in half-lives (34) and polymorphic expression of enzymes metabolizing NSAIDs (45) were possible causes. All of these could have contributed to the observed differences, although the exact mechanism is yet to be fully elucidated and experimental data on various NSAIDs are still lacking.
Liver Dose Adjustments
Meloxicam tablets are indicated for relief of the signs and symptoms of osteoarthritis see Clinical Studies (14.1). Drinking alcohol while taking meloxicam may increase the risk of gastrointestinal-related side effects or kidney damage. Serious skin reactions can occur during treatment with this medicine. Check with your doctor right away if you have blistering, peeling, or loosening of the skin, red skin lesions, severe acne or skin rash, sores or ulcers on the skin, or fever or chills while you or your child are using this medicine.
Weighted baseline characteristics for each NSAID treatment group are summarized in Table 1. The average age of participants was 55±17 years old, and 47% of them were men. Among these people, diclofenac (58%) was the most frequently prescribed NSAID, followed by naproxen (19%) and ibuprofen (10%). Unweighted baseline characteristics of subjects are listed in Supplemental Table 2. In adults, the maximum recommended daily oral dose of meloxicam is 15 mg regardless of formulation.
